  教育背景
  男,1955年5月生,博士,教授。2000年被聘為博士生導(dǎo)師
  中國動(dòng)物學(xué)會(huì)理事,中國海洋與湖沼學(xué)會(huì)理事,山東動(dòng)物學(xué)會(huì)常務(wù)副理事長,山東生化與分子生物學(xué)會(huì)副理事長。享受國務(wù)院政府特殊津貼。
  1981年山東師范大學(xué)本科畢業(yè)、1984年山東大學(xué)碩士研究生畢業(yè),留校工作。1998年獲博士學(xué)位。曾分別赴日本山口大學(xué)(1992)、九州工業(yè)大學(xué)(2000)、德國吉森大學(xué)(2002)等進(jìn)行合作研究。
  培養(yǎng)的博士生有1人獲山東省優(yōu)秀博士論文,多人獲山東大學(xué)校長獎(jiǎng)學(xué)金。研究生畢業(yè)后適合從事動(dòng)物學(xué)、水產(chǎn)科學(xué)、生化與分子生物學(xué)和生物工程等方面的教學(xué)與科研工作。
  
  研究方向
  研究領(lǐng)域?yàn)閯?dòng)物學(xué)、生化與分子生物學(xué)。研究方向?yàn)闊o脊椎動(dòng)物先天免疫,包括先天免疫的模式識別、信號轉(zhuǎn)導(dǎo)途徑和效應(yīng)分子(主要是抗菌肽)及其功能。講授無脊椎動(dòng)物天然免疫等課程。
  近十幾年來一直從事無脊椎動(dòng)物先天免疫研究。主持和參加國家自然科學(xué)基金、國家863、973、國家攻關(guān)和省部級等項(xiàng)目20余項(xiàng)。在The Journal of Immunology, Cellular and Molecular Life Sciences,Molecular Immunology, Developmental and Comparative immunology等國內(nèi)外刊物發(fā)表論文100余篇。獲國家發(fā)明專利5項(xiàng)。近年來在無脊椎動(dòng)物先天免疫研究方面取得了較好的進(jìn)展,獲教育部2010年高等學(xué)校科學(xué)研究優(yōu)秀成果獎(jiǎng)—自然科學(xué)一等獎(jiǎng)。 主要在下列幾個(gè)方面開展了較系統(tǒng)的工作。
  1.感染的模式識別研究:多種免疫識別受體的發(fā)現(xiàn),改變了先天免疫的早期的概念,先天免疫可以通過模式識別受體(PRRs)識別不同的病原相關(guān)分子模式(PAMPs),而實(shí)現(xiàn)對不同病原的識別,因而存在著某種特異性。我們以對蝦為材料,發(fā)現(xiàn)了多種模式識別受體分子,對它們的性質(zhì)和功能進(jìn)行了較系統(tǒng)的研究。包括革蘭氏陰性菌結(jié)合蛋白(Molecular Immunology,2007)、6種C-型凝集素(Molecular Immunology,2008, 2009,Developmental & Comparative Immunology, 2009; Fish & Shellfish Immunology,2009)等,發(fā)現(xiàn)它們具有結(jié)合不同病原菌或不同病原相關(guān)分子模式的功能。并提出對蝦圍食膜蛋白(Molecular Immunology,2006)和血小板反應(yīng)素(Molecular Immunology,2006)可能與免疫有關(guān),具有免疫識別的作用。
  2.免疫信號的調(diào)整與信號轉(zhuǎn)導(dǎo)途徑:在模式生物研究中,發(fā)現(xiàn)了多種信號轉(zhuǎn)導(dǎo)途徑與動(dòng)物的先天免疫有關(guān)。包括在Toll樣途徑、免疫缺陷信號轉(zhuǎn)導(dǎo)途徑(Imd)途徑、Jak-Stat途徑、轉(zhuǎn)化生長因子beta途徑、p38絲裂原活化的蛋白激酶途徑、細(xì)胞程序化死亡途徑和胰島素樣受體途徑等。我們近期的工作發(fā)現(xiàn)了多種可能參與免疫信號的調(diào)整的蛋白酶及其抑制因子,包括含有Clip結(jié)構(gòu)域的絲氨酸蛋白酶及其同系物(Fish & Shellfish Immunology, 2009)、絲氨酸蛋白酶抑制因子(Fish & Shellfish Immunol, 2009)。同樣發(fā)現(xiàn)了多種與免疫信號轉(zhuǎn)導(dǎo)相關(guān)的關(guān)鍵分子,包括Toll樣受體及其配體Sp?tzle (Fish & Shellfish Immunology,2009) 、IMD、STAT及多種磷酸激酶等相關(guān)的分子。初步實(shí)驗(yàn)結(jié)果顯示Toll途徑可能參與Crustin 抗菌肽表達(dá)的調(diào)控。
  3.免疫效應(yīng)分子的研究:無脊椎動(dòng)物主要的免疫效應(yīng)分子為抗菌肽(蛋白)。研究了中國對蝦和家蠅的多種抗菌肽的性質(zhì)、功能與表達(dá)調(diào)控,并成功進(jìn)行了原核和真核重組表達(dá)。首先對家蠅的抗菌肽進(jìn)行研究,克隆到了多種家蠅抗菌肽(包括天蠶素、防御素、攻擊素和溶菌酶等)基因,對其時(shí)空表達(dá)與調(diào)控進(jìn)行了研究,并進(jìn)行了原核和/或真核重組表達(dá),獲得了有抗菌活性的重組抗菌肽(Developmental & Comparative Immunology 2006, Cellular and Molecular Life Sciences 2006)。對蝦抗菌肽研究: 對中國明對蝦抗菌肽——對蝦素基因進(jìn)行了克隆與表達(dá)調(diào)控研究,得到2類(共4種)對蝦素基因,在對蝦中首次發(fā)現(xiàn)和命名對蝦素5,在大腸桿菌和酵母菌中成功表達(dá)了兩類對蝦素這些抗菌肽均顯示出廣譜抗菌活性,具有潛在的應(yīng)用前景(Fish & Shellfish Immunology 2004;Protein expression and purification 2004;Molecular Immunology,2007)。發(fā)現(xiàn)了多種含有WAP結(jié)構(gòu)域的抗菌肽,包括SWD(Aquaculture,2008)、甲殼肽(Fish & Shellfish Immunology,2010)、抗脂多糖因子、溶菌酶等,并進(jìn)行了重組表達(dá)的性質(zhì)研究,得到了有抗菌活性的上述分子。
  4.抗病毒機(jī)理的研究:RNA干擾途徑在無脊椎動(dòng)物中具有重要的抗病毒功能。我們在中國明對蝦中發(fā)現(xiàn)了轉(zhuǎn)錄激活反應(yīng)RNA結(jié)合蛋白(TRBP)與真核翻譯起始因子6(eIF6)之間存在著相互作用,可能與是RNAi途徑的重要組成成分(Journal of Immunology, 2009)。另外還發(fā)現(xiàn)了多種可能參與抗病毒反應(yīng)的蛋白(Molecular Biology Reports,2009)。
